Marie Curie EST position - Sensing Forum - ”Selective adsorbates for molecular recognition based om positively charged peptides”


Job Description: Marie Curie EST Sensing Forum position within ”Center for Protein Misfolding Disease, (CMD)”. 12-18 months early-stage resarch training at the division of Sensor Science and Molecular Physics at Linköpings universitet, Sweden.

The Marie Curie action within EU:s FP6 wants to stimulate the mobility of researchers, which implies that only non-Swedish citizens can apply for this announced position. The EST programme focuses om early stage researchers that are within their first four years of research training. The research within the Marie Curie EST Sensing Forum at Linköpings universitet, Sweden, stimulates a multidisciplinary approach to scientific questions within the sensing science area covering biology, chemistry, engineering, medicine and physics.

The largest membrane receptor family is the G-protein coupled receptor (GPCR). The adrenergic receptor (AR) is a sub - family within the GPCR family. This project is focused om design of surfaces with unique and specific properties. We will study the interaction between a G-protein specific peptide surface and G protein using Surface Plasmon Resonance (SPR). The peptide sequences are designed with the AR as a template, to investigate the activity and specificity of the molecular system. Peptide sequences and related molecular systems will be linked to gold surface by thiol chemistry. The, molecular system will be investigated by Ellipsometry, Atomic Force Microscopy, X-ray Photoelectron Spectroscopy, and Infrared Reflection Absorption Spectroscopy. The surface studies will be followed by protein interaction studies using Surface Plasmon Resonance, to investigate the activity and specificity of the molecular system.

One important issue is to study how the biomolecules are expressed when adsorbed to the surface. Co-adsorption of smaller inactive molecules as neighbours to the active molecule will be done, in order to make the active peptide sequences more available and to get the peptides to be expressed in a suitable way for protein interaction studies. Another way is to make a so called mini matrix to lift up the active biomolecule from the surface to get a flexibility in three dimensions, keeping the possibilities for analysing the molecular system by surface science techniques. We are looking for a collabolator with an interest of combining surface science and biological physics.

This position within the division of Sensor Science and Molecular Physics at Linköpings universitet is described as follows: ”Studies of the structural and functional properties of nanoparticles assemblies”. Main tasks: Development of metods and techniques for the assembly of nanoparticles into functional entities. Time plan. The proposed research and methodological developments will require 6-12 months of studies. The project will utilize protein folding as a general strategy to assemble nanoparticles into sandwich type structures on planar substrates or in homogeneous solution. The nanoparticles will be modified with biological recognition molecules (proteins) and analysed with respect to analyte binding using optical- and mass-sensitive biosensor technologies.

Time plan. The proposed research and methodological developments will require 6-12 months of studies. The project will utilize microcontact printing or photoactivation techniques to generate a molecular pattern (an array of spots) on gold and silicon substrates. The individual spots will be activated with recognition molecules (proteins) and analysed with respect to analyte binding using optical imaging techniques like surface plasmon resonance, ellipsometry or fluorescence microscopy.
